Experiment 5 (Assignment)

Electrochemical Determination of Glucose Concentration in Apple Juice

This assignment is a task of assisting a local producer to determine glucose content in their apple juice. The chemical measurements are required to be performed through electrochemical methods, which are known to be portable, easy to operate, fast and low cost in relative to other modern techniques such as HPLC, GC, Mass spectrometry etc. The electrochemical technique (e.g. Amperometry, Voltammetry, etc.) is covered in Chapter 17 and related information can also be found from Chapters 14 and 16.  Your responsibility includes:

  1. through comprehensive literature, search selecting a cost effective and robust electrochemical sensor,
  2. validating the electrochemical sensor and technique you just selected,
  3. preparing a list of chemicals and solution samples that are needed for the measurement,  4.  creating tables for recording the unknown sample and related responses, and

5.  discussing the reliability of the proposed measurements.

This assignment is to help students to comprehend and apply the topics that are discussed in Chapter 5 (both 8th and 9th edition).

Your laboratory report is expected to follow the same format as the previous laboratory reports:

General Layout

The report should be written in Arial, Times New Roman, Calibri, or similar font-type, with font sizes 11 or 12, and line spacing of 1.5 for all text.

Title Page

The first page of the report is the title page and must include the following information:

  1. Title of the experiment            3. Student’s name and ID number
  2. Date of submission     4. Lab section

Your final report must include the following 5 sections:

  1. Introduction

It is expected to have three parts (or paragraphs). The first part presents various electrochemical sensors that have been developed in the last two decades. This task can be accomplished through google search, searching key words “glucose” +

“electrochemical detection”. Alternatively, one can go to read several scientific journals on the subject about chemical sensing, such as Electrochemistry Communication, Electrochemical sensor B, etc. Part II of the introduction will present a particular electrochemical sensor you selected for this experiment and, importantly, clearly states why you choose this particular one (such as gold, or platinum nanoparticles,….) among all these existing glucose electrochemical sensors (e.g., due to its low cost? easy to fabricate? high sensitivity? robustness? …..). Part III presents the work reported in literature that deploys the sensor you just selected. This part can also have a brief description on the electrochemical techniques to be used, i.e., the principles of amperometry and voltametric measurements.

  1. Method validation

This section is expected to present and discuss experiment activities required to validate your method. One should refer to Chapter 5, in particular the section 2, when proposing the work activity.

  1. Chemical measurements

This section should contain the list of chemicals needed to complete the chemical measurements as well as procedures on the preparation of the required solutions. Upon the validation of your selected electrochemical method in section 2, which is assumed to show, hypothetically, a detection limit of 5.0 mg/L, and a linear range between 5.0 and 100.0 mg/L, you are expected to discuss the sample preparation on the basis that the apple juice sample may contain more than 200 mg/L glucose that is beyond the linear range of your detector. This section should also include tables that are ready to record unknown sample as well as their responses, similar to these tables seen in Experiments

1 to 4.

  1. Discussions and Conclusions

This section is meant to convey your understanding of the experiment. Discuss all important procedures and compare with those found in the literature used by other laboratories. When there are several sources of possible error, indicate to what extent each may have affected the final results. You may also suggest improvements or extensions to the experiment, and/or alternative methods of measurement.

  1. References

List these papers that are related to and assist you to complete this report. In the same manner as what you have done through Experiments 1 to 4, references should appear in the order they appear in the text. References in the text should appear as superscripted Arabic numerals.

Please upload the completed laboratory report to Blackboard in PDF format only by Monday, November 30 at 12 noon.
