Include background of microbes and microbiology, staining methods(preparation of smears and simple staining, negative staining, gram stain, acid fast stain, structural stains: endospore, capsule, flagella), also include microbial metabolism test(carbohydrate catabolism, fermentation, protein catabolism, and respiration)
